Membrane-Based Scanning Force Microscopy

چکیده

We report the development of a scanning force microscope based on an ultra-sensitive silicon nitride membrane transducer. Our is made possible by inverting standard geometry - in our instrument, substrate vibrating and tip at rest. present first topography images samples placed surface. measurements demonstrate that retains excellent sensitivity when loaded with presence tip. discuss prospects limitations instrument as quantum-limited sensor imaging tool.

Scanning force microscopy of artificial membranes.

Visualization of biological membranes by scanning force microscopy (SFM) has tremendously improved the current understanding of protein-lipid interactions under physiological conditions. SFM is the only tool to directly image processes on surfaces in aqueous solution at molecular resolution. Besides being a supportive means to confirm results on lipid phases and domains obtained from fluorescen...

DNA height in scanning force microscopy.

The measured height of DNA molecules adsorbed on a mica substrate by scanning probe microscopy is always less than the theoretical diameter. In this paper we show that, when imaged in ambient conditions, the molecules are usually immersed in the salt layer used to adsorb them to the substrate. This layer distorts the measurement of DNA height and is the main source of error but not the only one...

Combining Atomic Force Microscopy and Laser Scanning Confocal Microscopy

Introduction The visualisation of sample elements using various forms of microscopy relies not only on magnification but also contrast. As such, differing forms of microscopy offer different information about a sample. Laser scanning confocal microscopy (LSCM) provides information about the 3D location of a particular, labelled component within a sample and has the additional advantage of exclu...

Complementary visualization of mitotic barley chromatin by field-emission scanning electron microscopy and scanning force microscopy.

The surface structure of mitotic barley chromatin was studied by field-emission scanning electron microscopy (FESEM) and scanning force microscopy (SFM). Different stages of the cell cycle were accessible after a cell suspension was dropped onto a glass surface, chemical fixed, and critically point dried. Imaging was carried out with metal-coated specimen or uncoated specimen (only for SFM). Th...
